Epstein-Barr virus infects more than 95 percent of people, usually without symptoms. But sometimes its persistence in cells can lead to tumour formation. Now, researchers from Hong Kong and the UK have developed a fluorescing, molecular-sized probe, called L2P4, which can inhibit Epstein-Barr-related tumour growth while allowing researchers to see the targeted tumour cells. A team led by chemist Gary Ka-leung Wong from Hong Kong Baptist University developed the probe, which is formed of a nucleus-penetrating peptide linked to a fluorescent chemical compound. L2P4 targets a viral protein, called EBNA1, which is crucial for maintaining the Epstein-Barr virus (EBV) genome inside tumours. At a recent medical conference, scientist and Harvard professor George Church presented an argument causing anxiety among healthcare investors: The days of CRISPR gene editing might be numbered. • He called CRISPR a "blunt axe" and said "it's called editing, I think it's really genome vandalism." Church made the case that the biotechnology industry might be better off moving beyond cutting genomes and toward more precise gene-editing techniques. • In multiple recent speaking engagements, Church laid out four different gene-editing systems that may move the field "beyond cutting" for applications as diverse as industrial biotechnology, human medicine, and xenotransplantation. • Lambda-red recombinase: Allows direct modification of genomes resulting in fast turnaround times and few experimental steps. • Phage integrases: Simple method to make precise and controlled edits; also repairs sliced DNA without using cell's repair mechanisms. • TAL deaminases: The basis for base editing, which converts DNA base pairs in place without cutting the genome. • Chemical targeting: Engineering enzymes and proteins (including in the systems above) for direct genome modification without cutting DNA. • investors aren't yet concerned with precise gene editing of bacteria or plants, so those applications aren't top of mind for now, so we can set aside lambda-red and the use of recombineering for the time being. • Meanwhile, chemical targeting systems include a broad range of techniques that can be combined with other systems. • That leaves two competing technologies insofar as investors are concerned: phage integrase systems and base editing. • Both systems check more boxes on Church's list of characteristics for an ideal gene-editing system than CRISPR does. • Both are considered ideal candidates for in vivo and ex vivo. Importantly, both avoid triggering the problematic DNA repair mechanisms observed in some uses of CRISPR. • Base editing will provide the most immediate test. Loss-of-function mutation in one gene copy, termed haploinsufficiency, can lead to insufficient protein levels and result in human disease. • Matharu et al. tested whether a CRISPR-based activation system (CRISPRa) could rescue a haploinsufficient phenotype by increasing the gene expression levels of the existing normal copy. • By delivering this system into the mouse hypothalamus using adeno-associated virus, they rescued the obesity phenotype caused by haploinsufficiency of either of two genes known to promote obesity when mutated in mice and humans. • These results highlight the translational potential of the CRISPR activation system to treat haploinsufficient disease. Mesmo assim, é difícil entender. Explico: a manipulação de genes, em diversos contextos, pode auxiliar na supressão de fatores genéticos causadores de doenças. Assim, o desenvolvimento e o aprimoramento de ferramentas de edição de genomas possibilitam o surgimento de terapias focadas nos fatores de risco genéticos ao dano a que se deseja tratar, ou corrigir, ou até mesmo, abolir. É uma técnica molecular nova que permite editar o genoma (DNA). Portanto, esta técnica destaca-se devido ao seu fácil uso, alta especificidade, fácil manipulação 'in vitro' e 'in vivo', além da possibilidade da edição de múltiplos alvos simultaneamente. One of the main worries that the public has about CRISPR is that it could be used to create “designer babies” with increased levels of intelligence. • In the worst-case scenario, it could give an unfair advantage to parents who can afford to upgrade their offspring. Indeed, a Pew survey of US residents in July 2018 found their biggest worry was that CRISPR “will only be available to the wealthy.” • They needn’t worry, said three top gene-editing scientists this week. Intelligence is too complex to engineer and that’s a good thing, they say, since it “may ultimately save us from the hazards of humanity’s hubris." • The public’s concern about the creation of intellectual elites probably isn’t realistic, according an editorial published January 16 in the New England Journal of Medicine. • The article was written by George Daley, the dean of Harvard Medical School, and two European experts in embryology, Robin Lovell-Badge and Julie Steffann. • “In the long run, our greatest protection against inappropriate genome editing may be the implausibility of influencing traits such as intelligence, which emerge from complex interactions among multiple genes and environments,” they write. • We have linked more than a thousand genes to IQ or how far a person gets in school. Right now, scientists don’t know what those genes do, and what’s more, gene-editing tools aren’t yet up to the job of tailoring an embryo’s genes at so many locations. • To the editorial writers, that kind of ignorance is bliss. They are eager to see gene editing used to stamp out genetic disease in embryos—and they think it’s almost ready for the job, too. • Because serious congenital diseases like cystic fibrosis are often caused by specific DNA errors in single genes, it’s quite possible to repair them with CRISPR. Doing it in an embryo, they say, might be the best way to get rid of the problem. • Daley and other scientists now want to see this sort of medicine move forward. Baby Beta Cells Years of piling on the pounds and avoiding exercise are risky behaviours when it comes to developing diabetes. But a different and rare form of this disease can also strike at birth. Neonatal diabetes is sometimes caused by mutations in the insulin gene that affect the insulin-producing, beta cells of the pancreas. Researchers investigated further using stem cells created from people with these mutations. Using a genetic technique called CRISPR these mutations were corrected in some stem cells which were then grown in chemicals that turned them into beta-like cells (pictured), as confirmed by fluorescent microscopy of different markers for pancreatic cells. Analysing RNA molecules in both the mutant (top) and corrected (bottom) cells revealed that mutant cells were stressed and divided less. Following transplantation into mice, mutant cells weren’t any more likely to die than corrected cells but they did make less insulin, hinting at what goes wrong in neonatal diabetes. Wer ist der nächste Verwandte vom Mammut? Natürlich der #elefant . Und der hatte auf unserer Seite schon oft einen Gastauftritt. Aber das 3. Mal ist etwas ganz Besonderes: Eine abstrakte kleine Geschichte über das #wollhaarmammut , Steppengras und den Klimawandel. Schauplatz unserer Erzählung ist die sibirische Steppe. Denn ob ihr es wusstet oder nicht. In der #tundra Asiens liegen, begraben unter Permafrostböden, hunderte von gut erhaltenen Mammutknochen. Diese tauen aufgrund des Klimawandels immer weiter auf und legen diese riesigen Geschöpfe frei. Doch das Problem an abtauenden Permafrostböden ist, dass sie in sich gewaltige #CO2 Reserven speichern. Taut also der Boden auf, wird nach und nach eine gewaltige Menge an #kohlendioxid in die #atmosphäre geblasen. Aber was hat das Abtauen der Böden mit dem Wollhaarmammut zu tun? Das Vater-Sohn Gespann Sergei und Nikita Zimov hatten die Idee, die Tundra erneut mit mehr Leben zu erfüllen. Aufgrund der extremen Wetterbedingungen fällt es Lebewesen schwer, sich an dem Ort niederzulassen. Das Mammut hätte in der Kälte aber weniger Probleme. Der gute Nebeneffekt: Tiere fördern das Wachstum des Steppengrases. Diese hellen Gräser sind in der Lage, Sonnenlicht zu reflektieren und die Temperatur der Permafrostböden zu senken. Das wäre ein Erfolg für die ganze #welt . Und möglich macht’s die #wissenschaft . Durch Methoden wie #crispr können genetische Daten, die aus den Mammut-Skeletten entnommen und sequenziert, also gelesen wurden, in die #dns des asiatischen Elefanten eingefügt werden. Das #genom des asiatischen Elefanten ist zu 99% identisch mit dem des Wollhaarmammuts. Es müssen also Sequenzen gefunden werden, die sich zu denen des Elefanten unterscheiden oder gar nicht erst in der dns vorkommen. Diese neu erstellten Daten können dann einem Elefantenembryo eingesetzt werden. Und fertig ist die Wiederauferstehung des Mammuts. Obwohl es doch nicht ganz so einfach ist… Was haltet ihr davon? Young adults in China feel a powerful cultural obligation to marry and have kids, but that life plan suddenly looks unattainable to people told they’re infected with HIV, particularly for the many who can’t afford or are unaware of treatments that would allow them to have uninfected children. • So when an ambitious scientist offered HIV-positive men and their spouses what seemed to be a way out of this despair, several hundred couples in China jumped at the chance, expressing interest in a clinical trial that promised to deliver them babies forever protected from HIV infections. • The trial, involving gene editing of embryos created through IVF, led to the claimed birth of twin girls last November and a second, apparently still-ongoing, pregnancy. • The project has since been condemned by scientists worldwide as unethical human experimentation and Chinese authorities have halted the project while they investigate the scientist, He Jiankui. • Martin Yang, director of China AIDS Walk, told STAT that societal pressure to have children was likely an important factor — along with the parents’ desire to shield their children from the discrimination and ostracism people living with HIV endure in China. • Given these circumstances, Yang said, “there will always be a few hopefuls who want to try new technology.” [credit: Wendy Tang, STAT]🌐
