Scientists have found the Holy Grail of genetic engineering. Since its introduction four decades ago, genetic engineering has been a source of high hopes for health, agriculture, and industry. But it has also provoked deep anxiety, not least owing to the laborious nature of the genome-editing process. Now, a new technique, CRISPR-Cas, offers both precision and the ability to modify the genome text at several places simultaneously. But this has not eliminated reason for concern.... ....A mutation obtained by CRISPR-Cas, by contrast, is not distinguishable from a mutation that emerged spontaneously. That is why the US Food and Drug Administration has ruled that such constructs do not need to be labeled as genetically modified organisms. Previous techniques were especially arduous if one needed to modify several genes, because the process would need to occur sequentially. With CRISPR-Cas, the ability to perform genome modifications simultaneously has already enabled the creation of fungi and apples that do not oxidize, or turn brown, when they come into contact with air – a result that required several genes to be deactivated simultaneously. Ambitious efforts to prevent or beat cancer are important, but we can’t overlook or undervalue the incremental breakthroughs that could quickly reach patients and improve lives. • What if, for example, we used CRISPR to make chemotherapy a bit less terrible for patients suffering with cancer now? That’s something Eric Kmiec and his colleagues are trying to find out. • They are using CRISPR to disable an essential gene that is responsible for the development of chemotherapy resistance. To do this, they inject the CRISPR payload into a tumor using an FDA-approved viral delivery system. • Their experiments with the Wistar Institute have shown that the virus brings CRISPR to all the cells within the tumor - and no further. Once the payload gets inside cancer cells, it deactivates their self-defense mechanisms. • With this disruption, so the thinking goes, chemotherapy will be more effective because the gene or genes that confer resistance are disabled. • Put simply, if we disrupt a cancerous cells’ ability to defend themselves, we can use smaller doses of chemotherapy. That would make the treatment more effective, kill fewer healthy cells, and make the disruption to patients’ lives much more manageable. • The future of gene-editing is full of big things - big breakthroughs, big ethical questions, and big business - but we must not overlook the value of smaller wins. Scientists decoded the genome of rice in 2002. They completed the soybean genome in 2008. They mapped the maize genome in 2009. But only now has the long-awaited wheat genome been fully sequenced. • That delay says nothing about wheat’s importance. It is arguably the most critical crop in the world. It’s grown on more land than anything else. It provides humanity with a fifth of our calories. But it also has one of the most complex genomes known to science. • Using the completed genome, the team identified a long-elusive gene (with the super-catchy name of TraesCS3B01G608800) that affects the inner structure of wheat stems. • If plants have more copies of the gene, their stems are solid instead of hollow, which makes them resistant to drought and insect pests. By using a diagnostic test that counts the gene, breeders can now efficiently select for solid stems. • “It will take some time before the benefit to the breeding community is realized,” says Ravi Singh from the International Maize and Wheat Improvement Center in Mexico. • “But in our own program, we are already using this resource to identify important genomic regions behind traits like grain yield, disease resistance, tolerance to heat and drought, and nutritional quality.” • The IWGSC has also started to work out when different genes are turned on as wheat germinates and grows, and how these patterns of activity vary across the three subgenomes. • If scientists can figure out how to switch on specific genes at particular points in the plant’s life cycle, people could potentially breed wheat in real time, “in response to the growing season and environment,” Bentley says. “That would be incredibly cool.” • The IWGSC showed how easy it would also be to identify wheat genes that influence flowering time, and alter them with crispr to create varieties that bloom a few days earlier than usual. • Click the link in the bio to read about how scientists have found the genes in wheat that code for specific proteins responsible for disorders such as celiac disease and baker's asthma and more. An estimated 13 percent of Americans take antidepressant drugs for depression, anxiety, chronic pain or sleep problems. For the 14 million Americans who have clinical depression, roughly one third don’t find relief with antidepressants. • But now, researchers at Washington University School of Medicine in St. Louis and Sage Therapeutics in Boston are trying a different approach to alleviate depression. • Most antidepressant drugs target serotonin receptors, aiming to boost serotonin, a chemical thought to regulate mood and social behavior. Using CRISPR technology, the scientists have been able to target an altogether different type of receptor called delta-type GABA receptors. They think that natural mood-boosting substances in the brain can target these receptors. • “There’s a real need to develop more effective antidepressants,” said principal investigator Steven Mennerick a professor of psychiatry. • “The most commonly prescribed antidepressant drugs — such as Prozac, Paxil and Zoloft — were approved by the FDA more than 30 years ago, and there’s been a dearth of new antidepressants since then. A completely new approach is warranted.” • That new approach involves targeting GABA receptors with substances called neurosteroids, chemicals that occur naturally in the brain and are involved in emotional and motivational brain networks. • The research was conducted through the university’s Taylor Family Institute for Innovative Psychiatric Research, which was established in 2013 to develop new drugs for depression and other psychiatric illnesses. • “Neurosteroids are thought to selectively interact with delta-type receptors, and there’s evidence that those drugs may help patients suffering from depression,” said Mennerick. In a new study in cells, University of Illinois researchers have adapted CRISPR gene-editing technology to cause the cell's internal machinery to skip over a small portion of a gene when transcribing it into a template for protein building. • This gives researchers a way not only to eliminate a mutated gene sequence, but to influence how the gene is expressed and regulated. • Such targeted editing could one day be useful for treating genetic diseases caused by mutations in the genome, such as Duchenne's muscular dystrophy, Huntington's disease or some cancers. • The new CRISPR-SKIP technique, described in the journal Genome Biology, does not break the DNA strands but instead alters a single point in the targeted DNA sequence. • "We tested it in three different mammalian cell lines to demonstrate that it can be applied to different types of cells. We also demonstrated it in cancer cell lines because we wanted to show that we could target oncogenes." • "In Duchenne's muscular dystrophy, for example, just correcting 5 to 10 percent of the cells is enough to achieve a therapeutic benefit. With CRISPR-SKIP, we have seen modification rates of more than 20 to 30 percent in many of the cell lines we have studied." • Since the researchers saw some mutations at off-target sites, they are working to make CRISPR-SKIP even more efficient and specific. CRISPR: Revolutionary gene-editing tool explained! . . CRISPR has been hogging the scientific limelight in recent times. It is a powerful method of genetic manipulation, and scientists use it to remove, add or alter sections of DNA. It has the potential to treat several genetic diseases including cancer, and is also being used in agriculture. But how does it work? . . It is based off a natural system used by bacteria to protect themselves against viruses. Bacterial DNA has clustered regions of repeat sequences called CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats). Bacteria also have a protein called Cas9, which acts as "molecular scissors" ✂️ and can make targeted cuts in DNA. . . If a virus (phage) attacks bacteria, they destroy the phage’s DNA and store its fragments (called spacers) in between these repeat sequences. When the same phage attacks again, these spacer sequences synthesize a type of RNA called guide RNA (gRNA). This gRNA guides Cas9 to find complementary regions in the virus’s DNA. Cas9 then cuts this specific region in the viral DNA, and the phage can’t attack it again. . . Scientists have transformed the bacterial CRISPR-Cas9 system into a simple, programmable genome-editing tool. By artificially synthesizing gRNA, they direct Cas9 to induce targeted cuts in DNA in cells, and the cut allows them to make many modifications – for eg. delete a gene, correct defects (mutations) existing in these genes, or insert completely different genes in between. . . One month ago, the European Court of Justice decided that living organisms, whose genetic material was altered by modern genome editing techniques, have to be classified as genetically modified (GM). One of these techniques, that has become popular since its finding in 2012, is CRISPR-Cas. It has extensive potential for the prevention and treatment of diseases. Research with the help of CRISPR-Cas is performed on a variety of diseases, ranging from single-gene disorders to more complex conditions, including different cancers, HIV, mental and blood disorders. First cancer immunotherapy trials are already on their way. For those, patients' own immune T cells are extracted from blood and precisely manipulated via CRISPR-Cas to target cancer cells more efficiently. The T cells are then expanded in vitro (in a dish), before they are infused back into the patient. What exactly does CRISPR-Cas do? It acts as some sort of 'molecular scissors' to target locations in the genome precisely. The system consists of two parts. CRISPR is an RNA guide strand, that binds to a matching DNA sequence with high specificity, thereby targeting the enzyme Cas to a desired location. Once there, Cas modifies the DNA, e.g. by simply cutting it, such that the gene is silenced. The cells' repair mechanisms then come into play to restore the DNA helix, but the targeted gene will stay inactivated. Are there any risks? Even though CRISPR-Cas is the most precise genome editing tool up-to-date, recent studies have found a significant amount of off-target effects: that means the system can change the DNA at locations unintended, with unpredictable outcomes. When used for the modification of crops or live stock, this might have unintended consequences for the environment. Further, ethical concerns have been raised concerning the potential of choosing desirable human traits in the future.
